    An Extensible User Interface for Lean 4

    Contemporary proof assistants rely on complex automation and process libraries with millions of lines of code. At these scales, understanding the emergent interactions between components can be a serious challenge. One way of managing complexity, long established in informal practice, is through varying external representations. For instance, algebraic notation facilitates term-based reasoning whereas geometric diagrams invoke spatial intuition. Objects viewed one way become much simpler than when viewed differently. In contrast, modern general-purpose ITP systems usually only support limited, textual representations. Treating this as a problem of human-computer interaction, we aim to demonstrate that presentations - UI elements that store references to the objects they are displaying - are a fruitful way of thinking about ITP interface design. They allow us to make headway on two fronts - introspection of prover internals and support for diagrammatic reasoning. To this end we have built an extensible user interface for the Lean 4 prover with an associated ProofWidgets 4 library of presentation-based UI components. We demonstrate the system with several examples including type information popups, structured traces, contextual suggestions, a display for algebraic reasoning, and visualizations of red-black trees.     In den letzten Monaten stand das deutsche »Exportmodell« in der Kritik. Die deutschen Überschüsse seien mitverantwortlich für die Defizite der Nachbarländer und schaden den Handelspartnern. Ist diese Einschätzung gerechtfertigt? Für Gabriel Felbermayr, Universität Hohenheim, entbehren die Behauptungen einer soliden Grundlage. Dies bedeute nicht, dass die Ungleichgewichte nicht Anlass zu Sorge bieten, doch das fundamentale Problem liege in den Finanzmarktinstitutionen und nicht auf den Güter- oder Arbeitsmärkten. Nach Ansicht von Eckhard Janeba, Universität Mannheim, ist Deutschland zwar kein Vorwurf zu machen, da die maßvolle Haushaltspolitik den langfristigen konstitutionellen Vorgaben entspricht. Gleichwohl habe der deutsche Staat Politikentscheidungen getroffen, die sich in Handelsverzerrungen niederschlagen. Holger Görg, Universität Kiel, betrachtet die Frage von der Eben der Firmen aus. Eine relativ kleine Anzahl hochproduktiver Firmen zeichnet verantwortlich für einen Großteil der deutschen Exporte. Für Importeure gilt jedoch genau das Gleiche. Durch Importe kommt neue Technologie ins Importland, die von diesen einheimischen Firmen angewendet werden kann. Wenn nun beispielsweise Frankreichs Exporte nicht so sind, wie von Regierungsseite gewünscht, liegt das zu einem gewissen Teil mit an den beteiligten Unternehmen. Diese könnten durchaus von Importen profitieren, indem sie Zugang zu Technologie bekommen. Ansgar Belke, Universität Duisburg-Essen, verdeutlicht, dass die Exporterfolge Deutschlands auf Wettbewerbsvorteilen, die man sich durch Innovationen, technischen Fortschritt und eine moderate Lohnpolitik über Jahre hinweg erarbeitet hat, beruhen. Außerdem habe Deutschland auch die Rolle einer europäischen Konjunkturlokomotive ausgefüllt.     Effect of water activity in tortilla and its relationship on the acrylamide content after frying

    The objective of this study was to relate the tortilla minimum integral desorption entropy with acrylamide content during processing of tortilla chips. Tortilla pieces were stored at 30 °C at aw of 0.11-0.84 for 4 days and fried later in soybean oil at 180 °C for 25 s. The lowest acrylamide content was observed in tortilla chips made of non-stored tortilla (aw = 0.98) as well as in those prepared from tortilla stored in the minimum integral entropy (aw = 0.53). In addition, the color and texture values were similar in both cases. These results suggest that the reduction of the acrylamide content during processing of tortilla chips and other tortilla based foods thermally processed might be modified by factors such as moisture content, aw, and the physical state of water in the tortilla. Thus, the minimum integral entropy showed to be a reliable indicator to establish the most appropriate moisture conditions to obtain tortilla chips with reduced level of acrylamide when tortilla is dehydrated. © 2014 Elsevier Ltd.     T-cell epitopes of the major peach allergen, Pru p 3: Identification and differential T-cell response of peach-allergic and non-allergic subjects

    Lipid transfer proteins (LTPs), particularly peach Pru p 3, are the most relevant plant food allergens in the South of Europe, and, therefore, their allergic properties have been extensively studied. However, neither T-cell epitopes nor their effect on the patients’ T-cell response has been investigated in any member of the LTP panallergen family. The objective of the present study was to map the major T-cell epitopes of Pru p 3, as well as to evaluate their induced T-cell response in peach-allergic versus control subjects. Thus, peripheral blood mononuclear cells (PBMCs) from 18 peach-allergic patients and Pru p 3-specific T-cell lines (TCLs) from 9 of them were cultured with Pru p 3 and with a panel of 17 derived peptides (10-mer overlapping in 5 amino acids representing the full sequence of Pru p 3). Proliferation in 5-day assays was carried out via tritiated-thymidine incorporation, while IL4 and IFNγ production was assessed via sandwich enzyme-linked immunosorbent tests (ELISA) of TCL culture supernatants. The results were compared to those obtained from 10 non-peach allergic control volunteers. Two consecutive peptides showed the highest activation capacity. About 74% of PBMCs and TCLs recognized them, forming a single T-epitope: Pru p 365–80. Additionally, other specific T-cell epitopes were observed. Pru p 325–35 was detected by more than 60% of TCLs from peach-allergic patients, and Pru p 345–55 only activated PBMCs from control subjects. Interestingly, TCLs from patients were associated with a Th2-type, whereas control TCLs presented a Th1-type cytokine response. The major immunogenic T-cell epitope identified in Pru p 3, Pru p 365–80, is a good candidate to develop new vaccines for hypersensitivity reactions associated with LTP allergens from Rosaceae fruits

    Learning analytics to improve writing skills for young children – an holistic approach

    Purpose - Due to the important role of orthography in society, the project called IDeRBlog presented in this paper created a web-based tool to motivate pupils to write text as well as to read and to comment on texts written by fellow students. In addition, IDeRBlog aims to improve student’s German orthography skills and supports teachers and parents with training materials for their students. The paper aims to discuss these issues. Design/methodology/approach - With the aid of learning analytics, the submitted text is analyzed and special feedback is given to the students so that they can try to correct the misspelled words themselves. The teachers as well as the parents are benefiting from the analysis and exercises suggested by the system. Findings - A recent study showed the efficiency of the system in form of an improvement of the students’ orthographic skills. Over a period of four months 70 percent of the students achieved a significant reduction of their spelling mistakes. Originality/value - IDeRBlog is an innovative approach to improving orthography skills combining blogging and new media with writing and practice

    Area & Perimeter Surveillance in SAFEST using Sensors and the Internet of Things

    International audienceSAFEST is a project aiming to provide a comprehensive solution to ensure the safety and security of the general public and critical infrastructures. The approach of the project is to design a lightweight, distributed system using heterogeneous, networked sensors, able to aggregate the input of a wide variety of signals (e.g. camera, PIR, radar, magnetic, seismic, acoustic). The project aims for a proof-of-concept demonstration focusing on a concrete scenario: crowd monitoring, area and perimeter surveillance in an airport, realized with a prototype of the system, which must be deployable and foldable overnight, and leverage autoconfiguration based on wireless communications and Internet of Things. This paper reviews the progress towards reaching this goal, which is planned for 2015

    Community-developed checklists for publishing images and image analysis

    Images document scientific discoveries and are prevalent in modern biomedical research. Microscopy imaging in particular is currently undergoing rapid technological advancements. However for scientists wishing to publish the obtained images and image analyses results, there are to date no unified guidelines. Consequently, microscopy images and image data in publications may be unclear or difficult to interpret. Here we present community-developed checklists for preparing light microscopy images and image analysis for publications. These checklists offer authors, readers, and publishers key recommendations for image formatting and annotation, color selection, data availability, and for reporting image analysis workflows.     Omecamtiv mecarbil in chronic heart failure with reduced ejection fraction, GALACTIC‐HF: baseline characteristics and comparison with contemporary clinical trials

    Aims: The safety and efficacy of the novel selective cardiac myosin activator, omecamtiv mecarbil, in patients with heart failure with reduced ejection fraction (HFrEF) is tested in the Global Approach to Lowering Adverse Cardiac outcomes Through Improving Contractility in Heart Failure (GALACTIC‐HF) trial. Here we describe the baseline characteristics of participants in GALACTIC‐HF and how these compare with other contemporary trials. Methods and Results: Adults with established HFrEF, New York Heart Association functional class (NYHA) ≥ II, EF ≤35%, elevated natriuretic peptides and either current hospitalization for HF or history of hospitalization/ emergency department visit for HF within a year were randomized to either placebo or omecamtiv mecarbil (pharmacokinetic‐guided dosing: 25, 37.5 or 50 mg bid). 8256 patients [male (79%), non‐white (22%), mean age 65 years] were enrolled with a mean EF 27%, ischemic etiology in 54%, NYHA II 53% and III/IV 47%, and median NT‐proBNP 1971 pg/mL. HF therapies at baseline were among the most effectively employed in contemporary HF trials. GALACTIC‐HF randomized patients representative of recent HF registries and trials with substantial numbers of patients also having characteristics understudied in previous trials including more from North America (n = 1386), enrolled as inpatients (n = 2084), systolic blood pressure < 100 mmHg (n = 1127), estimated glomerular filtration rate < 30 mL/min/1.73 m2 (n = 528), and treated with sacubitril‐valsartan at baseline (n = 1594). Conclusions: GALACTIC‐HF enrolled a well‐treated, high‐risk population from both inpatient and outpatient settings, which will provide a definitive evaluation of the efficacy and safety of this novel therapy, as well as informing its potential future implementation

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
